MiMode™ PuraBead® HX2 is one of four resins in the hydrophobic mixed-mode range that harnesses both hydrophobic and hydrogen-bonding interactions to target proteins and viral particles where other approaches often fail. The hydrophobic family includes MiMode™ PuraBead® HX1, HX2, HL3, and HL4 resins, each tuned to offer slightly different binding profiles, allowing developers to identify the optimal solution for their molecule.

    Product Overview

    MiMode™ PuraBead® HX2 is a mixed-mode chromatography resin designed to provide a versatile and cost-effective solution for biomolecule purification. Built on the robust PuraBead® 6HF base matrix, it combines aromatic hydrophobic and hydrogen-bonding interactions, enabling both bind-and-elute and flow-through polishing applications.

    Unlike single-mode affinity solutions, MiMode™ PuraBead® HX2 offers broad selectivity, making it suitable as a platform technology across multiple molecule classes. The synthetic ligand ensures strong base stability, while the PuraBead® 6HF matrix delivers excellent flow properties and scalability from discovery through to large-scale manufacturing.

    With a wide operational pH range, compatibility with common buffer systems, and straightforward cleaning using sodium hydroxide, MiMode™ PuraBead® HX2 integrates seamlessly into existing workflows.     What is the pH stability of MiMode™ PuraBead® HX2?
    MiMode™ PuraBead® HX2 is stable in the long term (3 months) from pH 3 to pH 13. The recommended operating pH range for protein binding applications is pH 6.0 to pH 9.0, starting with a low pH elution step (5.0 – 3.0) with scope for further optimization.

    How should MiMode™ PuraBead® HX2 be stored?
    The recommended storage condition is 2 – 30°C in 20% ethanol.

    How is MiMode™ PuraBead® HX2 regenerated?
    Removal of any residual adsorbed material can be achieved by washing the column with 1.0 M NaOH. A contact time of 1 hour will normally suffice to ensure destruction of viable organisms, although up to 5 hours contact time may be required. No less than 5 column volumes are recommended. Once cleaning with NaOH is complete, wash with at least 3 bed volumes of equilibration buffer until the pH and conductivity of the column eluate is equal to that of the buffer entering the column. Complete this process prior to further use or storage in the storage buffer.
